Syed Afaq Hussain and Farhat Abbas Shah of the private Community Area Radio FM103 were arrested when they joined a protest over a cardiac facility.
The journalists had aired a programme critical of the running of the unit.
Information Minister Sheikh Rashid Ahmed recently launched an appeal for “responsible journalism”.
The journalists have applied for bail and a magistrate is expected to hear the case on Thursday.
The director of the radio station, Owais Bajwa, said one of the journalists was taken into custody at its offices and the other at a police station where he went to make inquiries about his colleague.
Mr Ahmed recently warned the Pakistani media to avoid sensational coverage of day-to-day events. Journalist organisations criticised the statement but have not yet commented on the latest arrests.
